Fisetin: a Powerful senotherapeutic


Fisetin is a bioactive flavonol molecule found in fruits and vegetables, such as strawberry, apple, persimmon, grape, onion, and cucumber. Several studies have reported fisetin to have neurotrophic, anticarcinogenic, and anti-inflammatory benefits. [1] Fisetin's anti-aging benefits are thought to be the result of its ability to eliminate cells that have undergone a process called senescence. Senescence is when a cell ages and permanently stops dividing but does not die. Over time, large numbers of these cells build up in tissues throughout the body and can release harmful substances that cause inflammation and damage to nearby healthy cells.

Several clinical studies are currently underway, including testing whether fisetin can improve physical function in breast cancer survivors, and in another study, comparing fisetin to the D+Q senolytic treatment (Dasatinib and Quercetin) in reducing frailty of adult survivors of childhood cancer. In a study that measured age via an epigenetic clock, combining D+Q with fisetin suggested that fisetin provides additional benefits beyond just senolytic activity, such as improved cognitive function and neuroprotection, and might be beneficial in modulating the effects of D+Q. [2]

A 2018 study [3] set out to test ten flavonoids for their senolytic properties. The study acknowledged that D+Q is a potent senolytic, improving numerous age-related conditions, including frailty, osteoporosis, and cardiovascular disease. However, this study aimed to identify natural flavonoids with even more potent senolytic activity.

The study examined critical connectivity tissue (human fibroblasts) that had become senescent from oxidative and genotoxic stress. They further tested the flavonoids on mice, evaluating age-related histopathology, disease markers, health span and lifespan. Of the 10 flavonoids tested, fisetin was the most potent senolytic. When they treated the old mice. Fisetin reduced senescent markers in multiple tissues, consistent with a hit-and-run senolytic treatment like D+Q. It was determined that Fisetin demonstrates senotherapeutic activity in both human and mice tissue. In mice, it was found that fisetin restored tissue homeostasis, reduced age-related pathology, and extended median and maximum lifespan.

It has been demonstrated that fisetin supports senescence by activating and regulating AMPK, MAPK, and mTOR pathways while also inhibiting CDK1, and CDK4 kinase enzymes. [4] A 2015 study indicated that AMPK activation can delay aging [5], while a 2019 study demonstrated mTOR inhibition has profound effects on life span and age-associated phenotypes across a wide array of organisms. [6]

mTOR is a protein kinase that controls cellular metabolism, catabolism, immune response, autophagy, survival, proliferation, and migration, to maintain cellular homeostasis. It is the target of the smart drug Rapamycin, the only compound known to extend mammalian lifespan.     You have a lot of great information here.

    I was taking the Bio-Fisetin from Life Extensions but didn’t notice anything. Then I tried the Renue Senolytique and really liked it, until I found Neuromergence which I feel is superior. Helped increase my energy, improved my sleep, and also gave me a noticeable boost in my sex drive. I also started on their fisetin. I take it in between neuromergence cycles, and it gives me more energy and focus.
